Das Wasserschloss Mespelbrunn liegt in der Gemeinde Mespelbrunn zwischen Frankfurt am Main und Würzburg in einem abgelegenen Seitental des Elsava-Tals im Spessart.
Schloss Mespelbrunn verdankt seine Entstehung einer Schenkung des Mainzer Erzbischofs Johann II. von Nassau. Dieser übereignete am 1. Mai 1412 seinem kurfürstlichen Forstmeister Hamann Echter den "Platz zum Espelborn", der im Talgrund an einem Weiher ein unbefestigtes Haus errichtete. Da der Spessart zu jener Zeit ein wilder und unerschlossener Wald war und Hussiten ihn als Zwischenstation für ihre Plünderungszüge nutzten, begann 1427 Hamann Echters gleichnamiger Sohn, aus dem unbefestigten Weiherhaus seines Vaters ein befestigtes Haus mit Mauern, Türmen und einem Wassergraben zu errichten (Forsthube).
In den folgenden friedlicheren Zeiten verwandelten die nachkommenden Generationen das abweisende Gemäuer in ein verträumtes Renaissanceschloss.
Sein heutiges Aussehen verdankt es größtenteils Umbauten zwischen 1551 und 1569, die Peter Echter von Mespelbrunn und seine Frau Gertraud von Adelsheim durchführen ließen. Über dem Turmportal des Nordflügels finden sich die Abbildungen beider und der Hausspruch:
Ehelich Lieb in Gott und stete Trew
Bringt Glück und Segen ohn alle Rew.
Mit Ernst und Fleis haben wir Gott vertraut,
Den Unseren zu Gut dies Haus gebaut.
