The only thing that comes to mind is perhaps the computer thinks its a model that doesn't have a transparency adaptor.

Epson Scan software?
Which mode are you using? If Full Auto Mode then try 'Customize' and change to film mode. Similar for Professional and Home modes (change document type).
How did you install the scanner when changing over computers? Scanner first and then software install? If so try the other way round (after removing all software).
You are removing the document mat? And unlocking the transportation lock?
Which film holders have you been trying? Placed correctly on scanner and nothing (dirt, Film ...) obstructing the initial sensor area (middle section near hinge)?

My V500 has developed this problem recently. I'll be mid-way through scanning negatives, then after a while (maybe an hour) the software will just quit on me when I hit the 'Preview' button, and then after restarting, refuses to select 'Film' mode.
Sometimes I can get it to select the film mode after a while of restarting the software, restarting the scanner, unplugging and plugging the USB cable in, fiddling with all the cables, but haven't found a reliable way of resetting the software. It's super annoying.
verablue, did you find a way of fixing it?

Did you fully unplug the connector for the lid and then plug it back in again before rebooting the scanner? You often can't just push on the plug to test it.

Yup, I fully unplugged it before switching back on again.

Had the same problem. Turning it off and unplugging the usb cable did the trick, but quite annoying. I have only had this scanner about a week.

I just had this happen and the answer I found is to reset the factory default settings on the software. With the software running, go to "configurations", then "other", then select restore factory settings. This took care of the problem for me. Hope it stays that way!

Download and buy the Vuescan software--it scans negatives better than the V500's software does.

Before installing third party software check this web page, I found it very useful when I first started using a V500. www.betterscanning.com/scanning/batchscanning.html

Has there been an update of some sort recently? I've had the V500 for months (and a 4490 for a year prior to that) and never had this problem happen until today.
edit: i reinstalled/factory reset, still no go.

i had the very same problem.
I did something similar to what some other flickr users have suggested, turn off scanner, unplug ALL the cables ( usb, power, cable betwen the scanner lib and unit ). Re-plug everything and start again.
that seem to work.

Unplugging then reattaching the cover from the base is what finally worked for me. I found that advice on Epson's US site under Top 10 support questions:
